Hepatotoxicity

Multiplexed Hepatotoxicity
Cell proliferation, Neutral lipid Accumulation, and Phospholipidosis

Our accelerated throughput hepatotoxicity multiplexed assay utilizes automated cell image acquisition and analysis technology to obtain simultaneous cellular cytotoxic mechanism data, High Content Cellular Assays.

MDS Pharma Services provides an effective in vitro cell-based screening model to assess human hepatotoxicity potential of drugs with multiplexed technologies examining a human hepatic cell line (HepG2) and measuring morphological and biochemical parameters at the single cell level.

Phospholipidosis and neutral lipid accumulation in liver cells are common and potentially dangerous side effects of pharmaceutical treatment which may also coincide with other toxicities. This assay provides essential data that can be used for prioritization and rank order of compounds in early discovery stage.

MDS Pharma Services Advantage
  • Access to automated, robust, state-of-the-art technology that maximizes data per assay by measuring critical markers of liver toxicity; Cell proliferation, Neutral Lipid accumulation, Phospholipidosis
  • Accelerated throughput screening (900 compounds per week)
  • Objective, consistent and fast
Service Features
  • Compound testing in pre-validated human HepG2 cells
  • Neutral lipid dye (LipidTox™ H34476)* - a marker of neutral lipid accumulation
  • Phospholipid analog (LipidTox™ H34350)* - a marker of phospholipidosis
  • Cell proliferation – relative cell count
  • Minimum compound requirements (2-3 mg) due to 384-well plate formats and non-contact acoustic energy based compound addition
  • A reference compound included as a control on each plate
  • 4 week turnaround time
